CARDIFF U. (UK) ? The body?s control mechanisms for delivering zinc to cells could be key to improving treatment for some types of aggressive breast cancer.

New research by Cardiff University and King?s College London has identified the switch that releases zinc into cells, with important implications for a number of diseases.

Zinc has long been known to play a vital part in human health. Too much zinc, or too little, can cause cell death. A growing body of evidence links zinc to disease states including neurodegeneration, inflammation, diabetes, and cancer.

Zinc levels in cells are controlled by protein molecules called zinc transporters. These move zinc in and out of the cell to ensure correct levels are maintained. Until now, scientists have not understood how the transporters release the zinc.

The research team has identified a switch, known as CK2, a protein that opens one transporter, called ZIP7, and allows the zinc to flow.

Earlier research by the team has already linked zinc delivery to types of breast cancer. Higher levels of intracellular zinc and the ZIP7 transporter were found in tamoxifen-resistant breast cancers. CK2 was also known to be more common in cancers which encourage cell growth.

As reported in the journal Science Signaling, the discovery that CK2 opens ZIP7 suggests that drugs that block this release of zinc could also block cancer development. Early results from clinical trials of CK2 inhibitors suggest they are performing well.

?We know that zinc, in the right quantities, is vital for development, our immune systems, and many other aspects of human health,? says Kathryn Taylor of Cardiff?s School of Pharmacy and Pharmaceutical Sciences. ?But when something goes wrong with the body?s zinc delivery system, it looks as though disease can result. In particular, our research has shown a link to highly aggressive forms of breast cancer.

Professor Christer Hogstrand from the Diabetes and Nutritional Sciences Division at King?s College London, says: ?Our discovery provides a mechanistic explanation for how the cell uses zinc to regulate different functions.

?The evidence that zinc is released on command in the cell and then regulates cellular processes seems to set it apart from other transition metals, such as copper and iron, in the way that it is used by the body. These findings should open the door for new research into the roles of zinc in health and disease.?

The research is funded by a Wellcome Trust University Research Award.

The role of pesticides is suspected, although not proven, in the occurrence of eight types of cancer on the rise for 25 years, according to the conclusions of the expert group ?Cancer and the Environment? of Inserm.
The collective expertise of the National Institute for Health and Medical Research (INSERM), funded by the French Agency of Health Safety Environment and Labour (Afsset), is the result of a thorough analysis of more than 1,800 scientific papers, made by a panel of experts from several disciplines (epidemiology, toxicology, clinical, occupational medicine, risk quantification). Made public Thursday, October 2 (1), it considered the link between environmental factors and nine types of cancer selected from the first part of the expertise in 2006, due to their increased incidence in 25 years: cancers lung, breast, thyroid, ovary, testis, prostate, mesothelioma, brain tumors and hematological malignancies (leukemia, lymphoma).

The number of cancers has increased in France over 90% between 1980 and 2005. Excluding the part due to demographic trends (growth and aging), this Mesothelioma increase in incidence remains high: 40% on average. ?The Hodgkin?s lymphoma increased by 6% a year became the third most common cancer in women,? said Guy Launoy, director of the Cancer and Population Unit Inserm. Inserm?s expertise was not limited to the evaluation of carcinogenic ?proven? like the study of the Academy of Medicine in collaboration with INSERM and the International Agency for Research against Cancer (IARC) (an offshoot of the WHO) in 2007 to cause a controversy, but also looked at environmental factors ?uncertain.?

According to the report, pesticides are suspected environmental factors for all cancers studied except mesothelioma. According to Jacqueline Clavel, a researcher in environmental epidemiology of cancers at Inserm, ?there is a strong suspicion on the role of carcinogenic pesticides.? These substances are thus one of the most suspected increase in brain tumors. However, the science points out that ?despite the observation of an increased incidence of several types of cancer (leukemia, lymphoma) in the agricultural population and in children associated with pesticide use, the link between cause and effect is not yet proven. ? Studies of causation must pursue this issue in order to eliminate another possible factor.

From INSERM, most studies suffer from high inaccuracy of exposure to pesticides (types of substances and use) and a lack of knowledge about the risks of environmental contamination (food , air, water). The expert makes recommendations to reduce exposure to pesticides and strengthen information from users and the monitoring of human exposure. The Movement for Rights and respect for future generations (MDRGF) expressed regret over a ?cautious to the extreme? and ?total lack of ambition in the recommendations it advocates.? MDRGF calls on Parliament to support measures to reduce 50% of pesticide use within 10 years and the promotion of organic farming in the voting laws Grenelle I and II this fall.

Other environmental factors ?suspects? for these nine cancers include radon, air pollution (fine particles), radio frequencies, artificial mineral fibers alternatives to asbestos, ionizing radiation, tobacco smoke and other pollutants (dioxins, PCBs, benzene).

The report also confirms the role of several environmental factors proved particularly asbestos for lung cancer and mesothelioma, or ionizing radiation for hematologic malignancies, lung cancer, brain tumors, breast cancer and thyroid.

The experts from INSERM stressed the importance of continuing research efforts to improve knowledge on the long-term effect of low doses of pollutants, including dioxins Thurs and ionizing radiation, as well as the impact of ?mixtures? of several factors and individual susceptibility to risk.

The rhetoric sounds so amazing and draws the American public in and says this is the dream and hope that Obama ran on. However, as you read through it and reviewing CNBC?s ?Obama?s Mortgage Refi Plan To Go Through FHA? that hope and dream fades quickly.

This proposal appears to have been written to in a way that it is either going to help essentially no one or be vetoed by the Republicans in Congress. Who in their right mind would use a tax on banks to pay for the changes that you are trying to force on the banks?

This just reads like a political stone wall for me, designed so President Obama can say ?I made an offer and the Republicans voted it down? on the campaign trail.

The only way to fix the housing situation is to get it out of Washington?s partisan politics and into the real world. But that?s only going to happen on a community-wide level and we just need Washington to stay out of the way.
